Liquid sporulation

1. Pick a single yeast colony and grow in 4 ml YPD medium overnight at 30°C.

2. Transfer 200 µl to 5 ml sterile ddH2O.

3. Centrifuge briefly to pellet the cells.

4. Resuspend cells in 2.5 ml SPM (0.3% Potassium acetate and 0.02% D(+)-Raffinose Pentahydrate).

5. Incubate at 25-30 °C for 2-5 days.

6. Check presence of asci with phase contrast microscopy and dissect.
