1. A genomic PCR can be used to determine the sex of a mouse embryo.
2. Smac primers:
Forward: 5'-TGAAGCTTTGGCTTTGAG-3'
Reverse: 5'-CCGCTGCCAAATTCTTTGG-3'
3. PCR products in male: 290 bp (Smac on Y) and 330 bp (Smac on X).
Females have only one product: 330 bp (Smac on X).
4. PCR program: 95 °C for 30 sec, 58 °C for 30 sec, 72 °C for 30 sec, 35 cycles.
