Endotoxin decontamination

Dry-heat

Endotoxin (pyrogen) may be decontaminated by heating at 250-260 °C in a depyrogenation oven (Sheldon Manufacturing) for no less than 1 hour. Lower temperature (180-200 °C) and longer time (3-4 hours) may also be used.

Acid or Base

Plastic and glassware may be decontaminated by soaking in strong acid (e.g., 0.1 M hydrochloric acid) or base (e.g., 0.5 M NaOH). Soak for 1 hour to overnight, and rinse thoroughly with pyrogen-free ddH2O.

Columns may also be decontaminated using strong acid (e.g., 0.1-1 M hydrochloric acid; 60% ethanol, 0.1-1 M acetic acid) or base (e.g., 0.1-2 M sodium hydroxide).

PyroCLEAN

Surfaces (e.g., tubing, rubber and metal), filtration columns and membranes may be decontaminated with PyroCLEAN (ALerCHEK, Cat. No. I30103).
