The following protocol is for the Quick Ligation Kit from BioLab.

Vector DNA ( 20-200 ng)
|
1 ml
|
|
Insert DNA ( 60-600 ng) ( 3 times vector)
|
1 ml
|
|
2X Quick Ligation Buffer
|
10 ml
|
|
Quick T4 Ligase
|
1 ml
|
|
ddH2O
|
7 ml
|
2. Mix and incubate the ligation at room temperature for 5 minutes.
3. Chill on ice. Don't heat-inactivate ligase. Transform or store at -20 °C.
