The low detection limit of Pico Green is ~ 1 ng/ml.

1. Make up a serial dilution of DNA standard (0-500 ng/ml).

2. Dilute the sample to concentrations between 50-400 ng/ml.

3. Mix 5 ml sample or standard, 1 ml Pico Green, and 94 ml TE in cuvettes (bigger volume may be needed) or a plate. Mix.

4. Read with a fluorometer.
