Cluturing of insect cells

Sf9 and Sf21 cells can be grown as monolayer cultures using flasks or tissue culture plates. Cells can be detached by pipetting the medium across the monolayer. No Trypsin-EDTA is necessary. These cells can be grown at room temperature or in a 27 °C incubator. They do not require CO2 incubator.

Sf9 and Sf21 cells can also be grown in suspension in a shaker or spinner. Seed cells at 0.5-1x 106 cells/ml in a shaker or a roller bottle. Cells transfered from monolayer to suspension may not grow well initially. Speed of shaking or spinning should increase gradually to adapt cells in the suspension culture.
