NaOH

1. Incubate the membrane in 0.4 M NaOH, 0.1% SDS at 25-65 °C for 30 minutes with gentle agitation.

2. Rinse in 0.2 M Tris-HCl pH 7.5, 0.1X SSC, 0.1% DSD for 30 minutes.

SDS

1a. Boil 0.5-1% SDS.

2a. Pour onto the membrane. Let cool to room temperature.

3a. Rinse in 0.2 M Tris-HCl pH 7.5, 0.1X SSC, 0.1% DSD for 30 minutes.

4. Proceed to hybridization or seal in a seal-a-meal bag and store at room temperature to -70 °C.
